Abstract

The lack of standardized demineralization protocols limits the bioactivity of dentin-derived biomaterials. Therefore, it is of interest to evaluate the effect of different durations of 12% EDTA demineralization on cryogenically nanogrinded dentin matrix (nDDM). Nanogrinded dentin samples were treated with 12% EDTA for 0, 2, 5, 10, 30 and 60 minutes and analyzed using Fourier Transform Infrared (FTIR) spectroscopy, while biological performance was assessed by ELISA, MTT assay and qRT-PCR in human dental pulp stem cells (hDPSCs). FTIR showed preservation of non-collagen associated amide bands and the 5-minute group exhibited the highest cytocompatibility, increased BMP-2 and TGF-β1 release and enhanced expression of DMP-1 and DSPP. Prolonged demineralization beyond 5 minutes resulted in reduced biological performance and alterations in matrix-associated spectral characteristics, indicating excessive demineralization. Thus, data shows that 12% EDTA demineralization for 5 minutes provides an optimal balance between mineral removal and non-collagenous matrix preservation, supporting the regenerative potential for vital pulp therapy.
