Abstract

Pentylenetetrazole-induced kindling causes progressive seizures, behavioural impairment and oxidative stress. Therefore, it is of interest to evaluate the effect of pyridoxine combined with low-dose levetiracetam in PTZ-kindled Wistar rats. The combination of pyridoxine 200 mg/kg and levetiracetam 50 mg/kg significantly reduced seizure severity, escape latency and malondialdehyde levels compared with PTZ controls (P < 0.0001). Combination therapy significantly increased glutathione, superoxide dismutase and catalase activities and produced the lowest median modified Racine score. Pyridoxine therefore potentiated the anticonvulsant and neuroprotective effects of low-dose levetiracetam during epileptogenesis.
